Background: HTLV I Tax is believed to activate viral gene expression by binding bZIP proteins (such as CRIB) and increasing their affinities for proviral THE target sites. Each 21 by THE target site contains an imperfect copy of the intrinsically bent CRE target site (the TRE core) surrounded by highly conserved flanking sequences. These flanking sequences are essential for maximal increases in DNA affinity and transactivation, but they are not, apparently, contacted by protein. Here we employ non-denaturing gel electrophoresis to evaluate TRE conformation in the presence and absence of bZIP proteins, and to explore the role of DNA conformation in viral transactivation.Results: Our results show that the TRE-1 flanking sequences modulate the structure and modestly increase the affinity of a CREB bZIP peptide for the TRE-1 core recognition sequence. These flanking sequences are also essential for a maximal increase in stability of the CREB-DNA complex in the presence of Tax.Conclusions: The CRE-like TRE core and the TRE flanking sequences are both essential for formation of stable CRIB-TRE-1 and Tax-CREB-TRE-1 complexes. These two DNA segments may have co-evolved into a unique structure capable of recognizing Tax and a bZIP protein.  相似文献

We prove several improved versions of Bohr’s inequality for the harmonic mappings of the form f=h+g¯, where h is bounded by 1 and |g(z)||h(z)|. The improvements are obtained along the lines of an earlier work of Kayumov and Ponnusamy, i.e. (Kayumov and Ponnusamy, 2018) for example a term related to the area of the image of the disk D(0,r) under the mapping f is considered. Our results are sharp. In addition, further improvements of the main results for certain special classes of harmonic mappings are provided.  相似文献

Significant increases in the measured elastic moduli with decreasing indentation depth have been previously found in various polymers by indentation tests with a Berkovich tip at micro-to nanometer length scales. These increases in the determined elastic moduli were related to second order displacement gradients which increase with decreasing depth when a conical tip is applied. When a spherical tip is applied, such depth dependence should not be present as the second order displacement gradients remain essentially unchanged with indentation depth. However, these gradients should be proportional to the radius of the spherical tip. To examine the notion of second order displacement gradient dependence in measurements of elastic moduli, indentation experiments are conducted on epoxy with spherical tips of different nominal radii. Accounting for tip imperfections, an increase in the determined elastic moduli is found with decreasing tip radius, which corroborates the notion of second order displacement gradient dependence.  相似文献

We incorporate the Boltzmann factors for inter‐monomer bending energy into the monomer growth direction choice in Rosenbluth's algorithm to model chains of arbitrary nearest‐neighbor rigidity. This allows for the consideration of compact (bent state lower in energy), free (straight and bent state equal in energy), or extended chains (bent state higher). We validate against, and compare to, various other results, showing very good agreement with known results for short chains and demonstrate the ability to model chains up to 500 segments long, far beyond the length at which the normal Rosenbluth method becomes unstable for reasonable nonzero bending energies. This approach is easily generalizable both to other energies determinable during chain growth, for example, polymers composed of more than one type of monomer with differing monomer interaction energies, as well as to other chain production algorithms. © 2019 Wiley Periodicals, Inc. 采用高阶无网格法求解薄板弯曲问题,在已发展的线性曲率光顺方案的基础上,通过引入泰勒展开技术,建立了能够精确再现纯弯曲和线性弯曲模式的节点积分方法。与之相比,目前无网格薄板分析主要采用的节点积分方法仅能精确再现纯弯曲模式。数值结果表明,本文方法可精确通过纯弯曲和线性弯曲试验,且能得到光滑、无振荡的弯矩场。与标准的高斯积分方法和目前已存在的节点积分方法相比,本文方法在计算精度、效率以及弯矩分布等方面均展现出显著优势。  相似文献

This paper investigates the essential conditions to improve the accuracy of a resistance spot welding computational study of advanced zinc coated steel sheets using rounded tip electrode. An experimental analysis is performed to highlight the required considerations for a suitable simulation. A sequential Electrical-Thermal-Metallurgical and Mechanical (ETMM) finite element analysis with appropriate precautions of the contact conditions enables to accurately simulate the nugget development during the welding. A critical smooth evolution of the contact radius is required. A fine meshing with an interfacial mesh size of at least 0.05 × 10−3 m combined with a coupling time step of 0.0025 s between the electrical-thermal-metallurgical and the mechanical analysis allows a regular incrementation of the contact radius, without burdening the time computing. Accurate values of the contact resistance depending on the interfacial pressure and temperature are essential for a good simulation of the nugget size. The ETMM calculation is successfully extended to the simulation of the welding of a typical two sheets assembly.  相似文献

基于台架应变测试试验方法,研究轿车车轮在疲劳试验中的应力状态。结果表明:车轮的径向疲劳试验时,轮胎会对车轮的载荷产生较大的影响,轮胎与转鼓的挤压变形以及正反转动都会对车轮的应力状态产生影响;车轮径向疲劳试验时,最大应变出现在轮辋与轮胎接触位置沿车轮圆周方向,而在靠近轮心位置的应变较小;车轮弯曲疲劳试验时,最大应变出现在轮辐靠近轮心的位置,最大应变出现在轮辐的长度方向;不同的载荷对车轮应变的变化规律并没有影响,但是会对最大和最小峰值产生影响。  相似文献

First, by using linear and trilinear estimates in Bourgain type analytic and Gevrey spaces, the local well‐posedness of the Cauchy problem for the modified Kawahara equation on the line is established for analytic initial data that can be extended as holomorphic functions in a strip around the x‐axis. Next we use this local result and a Gevrey approximate conservation law to prove that global solutions exist. Furthermore, we obtain explicit lower bounds for the radius of spatial analyticity given by , where can be taken arbitrarily small and c is a positive constant.  相似文献
